氙燈發(fā)出一束光,通過光調(diào)制器和UV鏡至濾光輪;濾過的光束進(jìn)入光導(dǎo)纖維,向上至測量裝置的透鏡;光束從蜂窩板培養(yǎng)孔的底部向上透過樣品懸浮液到達(dá)檢測器,檢測器得出濁度值后貯存于軟件系統(tǒng)中供用戶需要時調(diào)用。
測量裝置沿著Y軸移動,同時載有蜂窩板的培養(yǎng)箱盤沿著X軸移動,這樣就能在極短的時間內(nèi)快速測量蜂窩板中的所有培養(yǎng)孔的濁度值。

1、能夠排除培養(yǎng)基顏色變化對OD值的干擾
其他儀器通常在600 nm處測量菌懸液的濁度,但是600 nm處測量的不僅是濁度,而且某種程度上也包括培養(yǎng)液的顏色。Bioscreen采用寬波段濾光片技術(shù)專用于微生物學(xué)的濁度研究,因?yàn)槠浣Y(jié)果不會受到生長培養(yǎng)基顏色變化的影響,結(jié)果只表明濁度的生成狀況。
2、樣品不會產(chǎn)生冷凝或蒸發(fā)現(xiàn)象
技術(shù)確保蜂窩板蓋子上不產(chǎn)生液體冷凝現(xiàn)象,冷凝現(xiàn)象會使結(jié)果失真。蓋子也排除了樣品蒸發(fā)現(xiàn)象,因?yàn)檎舭l(fā)現(xiàn)象也會使結(jié)果失真。
3、***控溫使得結(jié)果可靠度高
溫度范圍為15.0~65.0℃(設(shè)置范圍:比環(huán)境溫度低9℃~高于環(huán)境溫度30℃),溫度變化精度可控制在±0.1℃。傳統(tǒng)的平板閱讀器不能保持這些允許變量,因此結(jié)果可靠度低。
4、光學(xué)測量的選擇
用戶能選擇2個波長另加1個寬波段(不受培養(yǎng)孔中顏色變化的影響)用于濁度測量。

測試舉例
全自動生長曲線分析儀是一個動態(tài)測量樣品濁度的光學(xué)機(jī)械儀器。要明白她是否能進(jìn)行一個特殊的測試,就有必要知道測試是否涉及培養(yǎng)液在生長期間的濁度(或顏色)變化。所有微生物(細(xì)菌、真菌、酵母和藻類)在培養(yǎng)液中繁殖都產(chǎn)生濁度。試管測試中凡是發(fā)生濁度和顏色變化的,大多數(shù)都能在她的“微測試試管"中來做。
除了多用途研究工作,全自動生長曲線分析儀也用于日常工作,如樣品中細(xì)菌數(shù)量的測量(食品質(zhì)量控制、細(xì)菌尿等)和微生物學(xué)測試(維生素、抗生素等化合物)。在很多其他研究中,如在測量抗生素對微生物的體外效果方面尤為突出。例如:
讓我們設(shè)想一下我們需要體外檢查5種新抗生素對于10種不同細(xì)菌的效果。為了比較,5種舊抗生素也用于測試。對于每種抗生素,我們將用5種不同的濃度;實(shí)驗(yàn)設(shè)置5個溫度處理和3個pH處理;并且所有實(shí)驗(yàn)都用3個平行樣。這樣,這種抗生素效果模型所需要的測試數(shù)為:

10×10×5×5×3×3=22500個生長曲線測試
用手工做這些測試(每天***多做20個測試)要花費(fèi)22500÷20=1125個工作日。假定每年有200個工作日,這樣就要花費(fèi)1125÷200=5.6年+資料處理時間。如果用Bioscreen C做生長曲線耗時少于12h(如果采用連續(xù)振蕩功能,這常能做到),每天就能做400個測試。Bioscreen C在56天內(nèi)可做22500個這樣的測試。這個實(shí)驗(yàn)用傳統(tǒng)的手工方法要耗時6年,而用Bioscreen C可在2個月內(nèi)完成。
